The nonrandomized MANHATTAN trial at Memorial Sloan Kettering Cancer Center (New York, NY) found that among patients with newly diagnosed multiple myeloma, the combination of weekly carfilzomib-lenalidomide-dexamethasone-daratumumab was associated with high rates of MRD negativity (71%). ORR rate was 100% and VPGR rate was 95%. The 1-year PFS was 98% and the 1-year OS was 100%. The therapy was deemed to be well tolerated.
