Venetoclax monotherapy in previously treated Waldenström macroglobulinemia
Dr. Jorge Castillo
BCL2 is overexpressed and confers prosurvival signaling in malignant lymphoplasmacytic cells in Waldenström macroglobulinemia (WM). Venetoclax is a potent BCL2 antagonist. A multicenter phase 2 study in the USA evaluated venetoclax monotherapy for previously treated WM. Venetoclax was safe and highly active, in patients with and without previous exposure to BTK inhibitors. CXCR4 mutation status did not affect treatment response.
