Given the inconclusive evidence of previous meta-analyses, Greek researchers performed an updated synthesis of the latest phase 3 studies of addition of CDK4/6 inhibitors to endocrine therapy (ET) for early-stage HR-positive, HER2-negative breast cancer. The meta-analysis included 4 RCTs (monarcE, NATALEE, PENELOPE-B, and PALLAS) with a total of17,749 patients. Addition of CDK4/6 inhibitors was associated with a significant improvement of invasive disease-free survival (HR 0.80; p=0.01) and a trend toward improved distant recurrence-free survival (HR 0.79; p=0.07), while the effect on overall survival remains inconclusive (HR 0.95; p=0.63).
