Tebentafusp versus investigator’s choice for previously untreated metastatic uveal melanoma

Metastatic uveal melanoma has a poor prognosis with a 1-year OS rate of about 50%. The multinational phase 3 study NCT03070392 evaluated the bispecific fusion protein tebentafusp (tebe) versus investigator’s choice (IC: pembrolizumab, ipilimumab, or dacarbazine) for HLA-A*02:01 + patients with previously untreated metastatic uveal melanoma. Tebe significantly prolonged overall survival compared to IC with estimated 1-year OS rate of 73,2% versus 57,5% (HR 0,51; p<0,0001). The OS benefit of tebe was seen in prespecified subgroups. The adverse event profile of tebe was manageable with a low rate of AE-related discontinuation.
