Sacituzumab govitecan (SG) is a antibody-drug conjugate directed against Trop-2. Systematic review of 38 published studies evaluated SG for triple-negative breast cancer (TNBC). Compared with chemotherapy, SG (10 mg/kg on days 1 and 8 of 21-days cycles) resulted in significant improvements in objective response rate, clinical benefit rate, progression-free survival, overall survival, and health-related quality of life. The benefit of SG over chemotherapy did not differ significantly by age, race, metastatic disease, number of prior lines of therapy, and presence of brain metastases. Compared with chemotherapy, SG was associated with a higher incidence of adverse events, especially neutropenia and diarrhea, which in some studies was associated with more frequent discontinuation of SG. The authors conclude that these results support the efficacy and safety of SG as treatment for TNBC.
