Systematic review of the literature and network meta-analysis found 20 eligible studies comparing anti PD-(L)1 agents as first-line therapy for locally advanced or metastatic non-small lung cancer (NSCLC) and 8 eligible studies comparing these agents as second- or later-line therapy. Tislelizumab with or without chemotherapy was more favorable than most comparator treatments and ranked as best or second-best treatment overall for progression-free survival in first-line squamous NSCLC, overall survival and progression-free survival in first-line non-squamous NSCLC with PD-L1 expression ≥ 50%, and overall survival and progression-free survival and grade ≥ 3 treatment-related adverse events in second- or later-line NSCLC of any histology. For overall survival and grade ≥ 3 treatment-related adverse events in first-line squamous NSCLC, tislelizumab with or without chemotherapy was comparable to other anti-PD-(L)1 therapies and combination therapies.
