The multinational phase 3 RELEVANCE trial randomised patients with previously untreated advanced follicular lymphoma (aFL) to lenalidomide plus rituximab (R2) or rituximab plus chemotherapy (R-chemo), followed by rituximab maintenance. The second interim analysis of the trial, after 6 years of follow-up, found no differences between the arms for the end points progression-free and overall survival. There were no new safety signals. The authors conclude that R2 followed by rituximab maintenance continues to demonstrate comparable and durable efficacy and safety, and represents an acceptable chemotherapy-free regimen for previously untreated aFL.
