Single-center phase 2 trial of loncastuximab tesirine plus rituximab for relapsed or refractory follicular lymphoma

A phase 2 trial at Sylvester Comprehensive Cancer Center (Miami, FL) evaluated the combination of loncastuximab tesirine and rituximab as second- or later line treatment for follicular lymphoma. The trial included 39 patients (median age 68 years; IQR 58-77) with an ECOG performance status 0-2. Patients received 3-week cycles of the combination. Primary end point was complete response rate at week 12. Complete response at week 12 was seen in 26 patients (CRR 67%). Serious treatment-emergent adverse events were seen in 4 patients (10%). There were no fatal TEAEs. The authors conclude that loncastuximab tesirine with rituximab showed clinically meaningful activity in relapsed or refractory follicular lymphoma, and had a maneagable safety profile.
