Single-center phase 2 trial of frontline acalabrutinib, lenalidomide, and rituximab for advanced stage follicular lymphoma with high tumor burden

A phase 2 trial at MD Anderson Cancer Center (Houston, TX) evaluated the combination of acalabrutinib, lenalidomide, and rituximab as first-line regimen for advanced follicular lympoma with high tumor burden. The most common grade 3-4 adverse events were neutropenia (58%) and liver function test elevation (17%). The best overall complete response rate was 92% and the best overall response rate was 100%. After median follow-up of 43 months, median progression-free survival and overall survival were not reached, with two-year PFS rate of 79% and two-year OS rate of 92%. The authors conclude that the combination of acalabrutinib, lenalidomide, and rituximab as first-line treatment for patients with advanced follicular lymphoma and high tumor burden was safe and effective.
