Single-center phase 2 trial of atezolizumab plus targeted therapy for anaplastic thyroid carcinoma

Anaplastic thyroid carcinoma (ATC) is a lethal cancer, with no approved therapies for non-BRAF mutated tumors. A nonrandomized phase 2 trial at MD Anderson Cancer Center (Houston, TX) evaluated the combination of atezolizumab and targeted therapy for ATC. Targeted therapy was selected based on the driver mutation: vemurafenib plus cobimetinib for BRAF V600E mutated tumors, cobimetinib for RAS or NF mutated tumors, and bevacizumab for non-BRAF/RAS/NF mutated tumors. The median overall survival of patients across these 3 cohorts was 19 months (95% CI 7.79-43.24), compared with historical median OS of 5 months. The authors conclude that atezolizumab plus genetically matched targeted therapy resulted in longer median overall survival than what was reported for historical controls.
