Single-center phase 1 trial of linperlisib plus chidamide for relapsed or refractory cutaneous T-cell lymphoma

Patients with relapsed or refractory cutaneous T-cell lymphoma (R/R CTCL) have limited treatment options. A phase 1 trial at Peking Union Medical College Hospital (Beijing, China) evaluated the combination of the PI3Kδ inhibitor linperlisib and the histone deacetylase inhibitor chidamide for R/R CTCL. This all-oral combination achieved an overall response rate of 59.1% (63.2% in mycosis fungoides and 33.3% in Sézary syndrome). Adverse events were mostly mild to moderate, with 22.7% of patients experiencing grade 3 events and no grade 4 or 5 events. The authors conclude that linperlisib plus chidamide showed promising efficacy and manageable toxicity, offering a potential new treatment option for advanced CTCL.
