Single-center phase 1 trial of GPRC5D-targeted CAR T-cell therapy for relapsed or refractory multiple myeloma

CT071 is an autologous CAR T-cell therapy directed against GPRC5D. A phase 1 trial at Shanghai Changzheng Hospital (China) evaluated CT071 for relapsed or refractory multiple myeloma after at least 3 previous lines of therapy. No dose-limiting toxicities were observed, and the recommended phase 2 dose was determined at 0.1 x 10⁶ CAR T cells per kg. No treatment-related deaths occurred. The objective response rate was 100% with a complete response or better rate of 50%. The authors conclude that CT071 demonstrated an encouraging safety profile with compelling activity in patients with relapsed or refractory multiple myeloma.
