Single-center phase 1-2a study of bispecific CS1-BCMA CAR-T cells for relapsed or refractory multiple myeloma

Multiple myeloma is a heterogeneous disease that poses a challenge for single-target immunotherapies. Researchers at Huazhong University of Science and Technology (Wuhan, China) constructed bispecific CS1-BCMA CAR-T cells aiming to augment BCMA targeting with CS1 for relapsed or refractory multiple myeloma (RRMM). At median follow-up of 246 days, 81% of patients had and overall response and attained minimal residual disease-negativity, and 38% reached a stringent complete response. Among the responders, 1-year progression-free survival and overall survival rates were 56% and 73%, respectively. Cytokine release syndrome was seen in 38% (grade 1-2 in 31%), while no neurotoxicity was observed. The authors conclude that CS1-BCMA CAR-T cells for RRMM were clinically active with good safety profiles.
