BMS-986156 is an agonist of the glucocorticoid-induced tumor necrosis factor receptor (TNFR)-related protein and promotes increased effector T-cell activation. A phase 1-2 study at MD Anderson Cancer Center (Houston, TX) evaluated BMS-986156 in combination with ipilimumab or nivolumab with or without stereotactic ablative radiotherapy (SABR) among patients with stage IV solid malignancies resistant to standard treatments. The treatment was tolerated, with 10% of patients discontinuing due to treatment-related adverse events, 44% experiencing grade 1-3 TRAEs, and no patients with grade 4 or 5 TRAEs. Among the patients who were evaluable for efficacy, 48.7% had stable disease and 7.7% had partial response Abscopal disease control rate was 38.5% and abscopal response rate was 7.7%. The authors conclude that BMS-986156 in combination with ipilimumab or nivolumab with or without SABR was well-tolerated and had encouraging activity among patients with advanced solid tumors resistant to standard treatments.
