The Drug Rediscovery Protocol (DRUP) evaluates off-label registered targeted treatments in patients with an actionable molecular profile. DRUP results of crizotinib for MET-mutated advanced non-small cell lung cancer have been published. Among 24 response-evaluable patients after completing at least one full treatment cycle, two patients achieve complete response, 13 partial response, and two stable disease during at least 16 weeks, for a clinical benefit rate of 70.8% (95% CI 48.9-87.4). The median duration of response was 9.3 months (95% CI 6.5-NE), the median progression-free survival was 10.2 months (6.0-20.1), and the median overall survival was 13.0 months (9.0-NE). Twelve of 30 patients (40%) had 23 grade 3 or higher treatment-related adverse events, leading to treatment discontinuation in three patients (10%). The authors conclude that crizotinib is a valuable treatment option for MET-mutated advanced non-small cell lung cancer.
