The multinational phase 3 ASCENT trial randomized patients with previously treated metastatic triple-negative breast cancer (mTNBC) 1:1 to the anti-Trop-2 antibody drug conjugate sacituzumab govitecan (SG) or single-agent chemotherapy of physician’s choice (TPC). Final analysis of the trial found that SG versus TPC was associated with superior progression-free and overall survival. SG improved PFS over TPC in each Trop-2 expression quartile; a trend was observed for improved OS across quartiles. SG had a manageable safety profile, with no treatment-related deaths. The safety profile was consistent across all subgroups. The authors conclude that these data confirm the clinical benefit of SG over chemotherapy, reinforcing SG as an effective treatment option in patients with mTNBC in the second line or later.
