An investigator-initiated, single-arm phase 2 study at the University of Pennsylvania (Philadelphia) evaluated rucaparib maintenance therapy for platinum-sensitive advanced pancreatic cancer with germline or somatic pathogenic variant in BRCA1, BRCA2, or PALB2. Rucaparib maintenance was a safe and effective therapy. Responses occurred in 11 of 27 patients with gBRCA2, 3 of 6 patients with gPALB2, and 1 of 2 patients with sBRCA2. The median duration of response was 17.3 months, the median progression-free survival was 13.1 months, and the median overall survival was 23.5 months.
