Retrospective study of histologic and immunologic factors associated with response of advanced sarcoma to immune checkpoint inhibitors

A retrospective study at Stanford University (Palo Alto, CA) investigated histologic and immunologic factors associated with the response of advanced sarcoma to immune checkpoint inhibitors (ICIs). Objective response rate and progression-free survival were highly variable across sarcoma histological subtypes. Highest ORR to ICIs was seen in Kaposi sarcoma, alveolar soft part sarcoma, and angiosarcoma, whereas the ORR was 0% for osteosarcoma and synovial sarcoma. Median PFS was not reached in Kaposi sarcoma en alveolar soft part sarcoma. The ORR for sarcomas with PD-L1 ≥ 1% was 27.8% while the ORR for sarcomas with tumor mutational burden ≥ 10 mutation per megabase was 28.6%. The authors conclude that these findings provide further insight into the sarcoma histologic and immunologic factors that correspond with response tot ICIs.
