Amivantamab is a bispecific anti-EGFR-MET antibody. The retrospective Amexon 20 GFPC study in France evaluated amivantamab for EGFR exon-20-mutant non-small cell lung cancer in heavily pretreated patients. Partial response was seen in 35%. Among patients with brain metastasis, 23% had partial response and 69% had disease control. Median PFS was 3.5 months (95% CI 2.6-5.8) and median OS was 11.3 months (8.0-17.8). Dose reduction was required in 17.0% and permanent discontinuation due to adverse events in 10.3%. The authors conclude that amivantamab demonstrated clinical activity in EGFR exon-20-mutant NSCLC, including in patients with brain metastasis, with a manageable safety profile.
