There are only limited real-world data comparing first-line palbociclib plus aromatase inhibitor (AI) versus AI alone for patients with HR-positive, HER2-negative metastatic breast cancer (mBC). Analysis of the Flatiron Health Analytic Database found median time to chemotherapy 37.4 months with first-line palbociclib plus AI versus 29.2 months with AI alone (HR 0.77; p<0.0001) and median real-world progression-free survival with second-line therapy 32.6 months versus 20.7 months (HR 0.62; p<0.0001). The authors conclude that this real-world analysis supports the use of palbociclib plus an AI as first-line treatment for HR-positive, HER2-negative mBC (visual abstract).
