Randomized study of pegasparaginase-based versus L-asparaginase-based regimen for newly diagnosed advanced ENKL

The L-asparaginase based SMILE regimen ((dexamethasone, methotrexate, ifosfamide, L-asparaginase, and etoposide) has shown efficacy for extranodal natural killer/T-cell lymphoma (ENKL). However, the safety profile was not favorable. A randomized study in China compared SMILE versus DDGP (dexamethasone, cisplatin, gemcitabine, and pegasparagase) for newly diagnosed advanced ENKL. DDGP was associated with better PFS and OS and less hematologic toxic effects (visual abstract).
