Targeted inhibition of the PD-L1/PD-1 pathway might be further amplified through combination with anti-TIGIT immune checkpoint inhibitors, such as tiragolumab. The multinational randomized phase 2 study CITYSCAPE evaluated addition of tiragolumab to first-line atezolizumab for recurrent or metastatic NSCLC with tumor proportion score 1% or higher. Tiragolumab plus atezolizumab was well tolerated, with a safety profile generally similar to that of atezolizumab alone. Addition of tiragolumab to atezolizumab resulted in a clinically meaningful improvement of objective response rate and progression-free survival.
