Pooled analysis of two multinational phase 1-2 trial of garsorasib for advanced pancreatic cancer with KRAS G12C mutation

Garsorasib (D-1553) is a selective oral KRASG12C inhibitor with activity in NSCLC and CRC. Pooled analysis of two multinatonal phase 1-2 trials evaluated garsoraib 600 mg twice daily for KRASG12C-mutated advanced pancreatic cancer. The analysis included 24 patients with a median follow-up of 8.9 months (range 1.1-22.9). Among 22 evaluable patients , the confirmed objective response rate was 45.5% (95% CI 24.4-67.8) with a median duration of response of 6.4 months (4.2-16.4). The median progression-free survival was 7.6 months (95% CI 3.3-8.5) and the 6-months overall survival rate was 79.2% (57.0-90.8). Treatment-related adverse events occurred in 75% of patients; grade ≥ 3 events in 25%. No TRAEs led to treatment discontinuation. The author conclude that garsorasib demonstrated encouraging antitumor activity and a tolerable safety profile among patients with KRASG12C-mutated advanced pancreatic cancer.
