Pooled analysis of rPFS and cPFS as surrogates for OS in metastatic hormone-sensitive prostate cancer
Dr. Susan Halabi
Despite major advances in the treatment of metastatic hormone-sensitive prostate cancer (mHSPC), most patients with die of the disease. Phase III trials assessing new therapies in mHSPC with overall survival as the primary end point will take approximately a decade to complete. Pooled analysis of individual patient data of 9 randomized trials found that the surrogate threshold effect of radiographic progression-free survival for overall survival was 0.80, while the surrogate threshold effect of clinical progression-free survival for overall survival was 0.81. The authors conclude that both rPFS and cPFS appear to be promising surrogate end points for OS.
