Larotrectinib is a highly selective and CNS-active tropomyosin receptor kinase (TRK) inhibitor, with demonstrated efficacy across TRK fusion-positive cancers, regardless of tumor type. Pooled analysis of two clinical trials found that larotrectinib was highly active in patients with advanced TRK fusion-positive lung cancer, including those with CNS metastases, with rapid and durable responses, extended survival benefit, and a favorable long-term safety profile.
