Phase 2 trial of sequential CD19 and CD22 CAR T-cell therapy for childhood refractory or relapsed B-cell acute lymphocytic leukemia

Relapses frequently occur following CD19-directed chimeric antigen receptor (CAR) T-cell therapy for relapsed of refractory B-cell acute lymphocytic leukemia (R/R B-ALL) in children. A phase 2 trial at Beijing GoBroad Boren Hospital evaluated CD19-directed CAR T-cell therapy, followed by CD22-directed CAR T-cell therapy after reaching minimal residual disease-negative complete remission. This sequential strategy induced deep and sustained responses with an acceptable toxicity profile. The authors conclude that this strategy potentially may provide long-term benefits for children with R/R B-ALL.
