G protein coupled receptor, class C group 5 member D (GPRC5D) is primarily expressed on malignant plasma cells in multiple myeloma, while normal tissue expression is restricted to the hair follicle. GPRC5D is considered to be a promising surface target for multiple myeloma immunotherapy. A phase 2 trial at Xuzhou Medical University (China) evaluated anti-GPRC5D CAR T cell therapy for relapsed or refractory multiple myeloma (RRMM). At a median follow-up of 5.2 months, the overall response rate was 91% (95% CI 76 to 98), including stringent complete response in 33% and complete response in 30%. Response was seen in all patients with prior anti-BCMA CAR T-cell therapy. The safety profile was manageable, with no grade 3 or higher cytokine release syndrome. Anti-GPRC5D CAR T-cell treatment is another effective CAR-T cell option for RRMM, especially in patients previously treated with anti-BCMA immunotherapy.
