Mesotheliomas frequently harbor loss of the chromosome 9p21.3 locus, which is associated with loss of the tumor suppressor p16ink4A, an endogenous suppressor of CDK4 and CDK6. Genetic restoration of p16ink4A suppresses mesothelioma in preclinical models. The phase 2 MiST2 trial in the UK evaluated abemaciclib for p16ink4A-deficient mesothelioma previously treated with chemotherapy. The study met its primary end point, with disease control at 12 weeks in 14 (54%) of 26 patients (95% CI 36-71).
