Phase 2 study of pegylated liposomal doxorubicin for heavily pretreated HER2-negative metastatic breast cancer

A study at Peking University Cancer Hospital & Institute (Beijing, China) evaluated pegylated doxorubicin (PLD) for HER2-negative metastatic breast cancer (mBC) in patients who had received two to five earlier lines of anthracycline and taxanes. In total, 59.1% of patients had 3 or more metastatic sites, 86.4% had visceral disease, and 63.8% had liver metastases. With the PLD product Duomeisu® 40 mg/m2 every four weeks, the median PFS and OS were 3.7 months (95% CI 3.3-4.1) and 15.0 months (12.1-17.9), respectively. The most common grade 3 adverse events were neutropenia (7.3% of patients) and fatigue (4.9%). One patient had LVEF decline of 11.4% from baseline after five cycles of PLD therapy. The authors conclude that Duomeisu® was effective and well tolerated in patients with heavily pretreated HER2-negative mBC.
