Phase 2 study of osimertinib plus savolitinib after first-line osimertinib for EGFR-mutated advanced NSCLC with MET alterations

The multinational phase 2 ORCHARD study was conducted to identify optimal treatments following progressive disease (PD) of advanced NSCLC on first-line osimertinib. The results of the osimertinib plus savolitinib module have now been published. Thirty-two patients were enrolled; all had tumors with MET amplifcation. Confirmed objective response rate on osimertinib plus savolitinib was 47%; median duration of response was 14.5 months; median progression-free survival was 7.6 months. Fourteen patients (44%) had grade 3 or higher treatment-emergent adverse events. Median overall survival was 20.7 months. The authors conclude that osimertinib plus savolitinib demonstrated encouraging clinical benefit in patients with EGFR-mutated advanced NSCLC and MET amplification following PD on first-line osimertinib. Safety was consistent with profiles of the individual drugs.
