Phase 2 study of cobimetinib plus vemurafenib for colorectal cancer with BRAF mutations

The Targeted Agent and Profiling Utilization Registry (TAPUR) study aims to evaluate the antitumor activity of FDA-approved drugs outside their approved indications in patients with advanced cancers. One of the TAPUR cohorts consists of patients with advanced CRC with BRAF mutations treated with the BRAF inhibitor vemurafenib plus the MEK inhibitor cobimetinib. Among 27 evaluable patients, the DCR and ORR were 52% (95% CI 35-65) and 30% (95% CI 14-50), respectively. Thirteen patients had at least one grade 3 adverse event or serious adverse even possibly related to the treatment. The authors conclude that the combination of cobimetinib and vemurafenib had antitumor activity in heavily pretreated patients with CRC with BRAF mutations.
