Phase 1b-2 study of ciltacabtagene autoleucel for relapsed or refractory multiple myeloma

The multicenter phase 1b-2 CARTITUDE-1 study evaluated the BCMA-directed CAR T-cell therapy ciltacabtagene autoleucel in patients with heavily pretreated (median 6 previous lines of therapy) multiple myeloma. The ORR in patients treated at the recommended phase 2 dose was 97%. Time to first response was 1 month; responses deepened over time. With median follow-up of 12.4 months, median duration of response was not reached. PFS rate at one year was 77% and OS rate at one year was 89%. The safety profile was manageable.
