There is an unmet need for more efficacious therapies for patients with metastatic colorectal cancer with BRAF-V600E mutation. The phase 1b-2 EVICT trial (‘erlotinib and vemurafenib in combination trial’) evaluated BRAF and EGFR inhibition for mCRC (n=32) and other metastatic cancers (n=7) with this mutation. No dose-limiting toxicities were observed. Overall response rates were 32% in the mCRC cohort (clinical benefit rate 65%) and 43% in the other cancers cohort (CBR 100%). The authors conclude that EVICT demonstrated a safe and novel combination of two oral inhibitors targeting BRAF and EGFR.
