The phase 1-2a NCT Neuro Master Match umbrella trial at the University of Heidelberg (Germany) evaluated various matched targeted treatments for newly diagnosed glioblastoma:alectinib, idasanutlin, palbociclib, vismodegib, and temsirolimus. All patients received radiotherapy. The temsirolimus subtrial demonstrated a PFS at 6 months of 39.1% and median overall survival of 15.4 months in patients with activited mTOR signaling. The authors conclude that these results support further investigation of temsirolimus plus radiotherapy for glioblastoma with activated mTOR signaling.
