Patient-reported outcomes of the phase III HIMALAYA study of tremelimumab plus durvalumab for unresectable hepatocellular carcinoma

In the multinational phase 3 HIMALAYA study of patients with unresectable hepatocellular carcinoma, previously untreated patients were randomized 1:1:1 to single tremelimumab regular interval durvalumab (STRIDE), sorafenib, or durvalumab monotherapy. STRIDE significantly improved overall survival versus sorafenib, and durvalumab monotherapy was noninferior to sorafenib. Patient-reported outcomes have now been published. Time to deterioration in GHS/QOL, functioning, and disease-related symptoms was longer with STRIDE versus sorafenib, and the likelihood of clinically meaningful improvement was greater with STRIDE versus sorafenib. In general, STRIDE and durvalumab regimes were associated with similar times to deterioration and similar improvements for PROs. The authors conclude that these data lend further support to STRIDE as an option for the frontline treatment of advanced hepatocellular carcinoma.
