Overall survival with xevinapant plus chemoradiotherapy versus placebo plus chemoradiotherapy for LA HNSCC

Xevinapant is an orally available antagonist of inhibitor of apoptosis proteins (IAPs) with the potential to enhance the antitumor activity of cisplatin and radiotherapy. A phase 2 study at 19 centers in France and Switzerland evaluated addition of xevinapant to cisplatin-based chemoradiotherapy for unresected locally advanced head and neck squamous cell carcinoma. Addition of xevinapant to CRT was associated with prolongation of the duration of response and improvement of five-year OS (median, not reached versus 36.1 months; aHR 0.47; p=0.0101).
