Patients with HER2-positive metastatic breast cancer who have disease progression after therapy with multiple HER2-targeted agents have limited treatment options. Tucatinib is an investigational, oral, highly selective inhibitor of het HER2 tyrosine kinase. Final overall survival analysis of the multinational phase 3 HER2CLIMB trial found that tucatinib compared with placebo added to trastuzumab plus capecitabine resulted in clinically meaningful survival benefit for patients with pretreated HER2-positive metastatic breast cancer, including patients with brain metastases. The tucatinib combination was well-tolerated with a low rate of discontinuation due to adverse events (5.9%).
