Overall survival with trastuzumab deruxtecan versus trastuzumab emtansine for HER2-positive metastatic breast cancer
Prof. Sara Hurvitz
The multinational phase 3 DESTINY-Breast03 trial included patients with progressive HER2-positive metastatic breast cancer after at least one line of anti-HER2 therapy. Patients were randomized to trastuzumab deruxtecan (T-DXd) or trastuzumab emtansine (T-DM1). Results of the study presented at the San Antonio Breast Cancer Symposium showed that the overall survival was significantly better with T-DXd than with T-DM1 (HR 0.64; p=0.0037). Updated median progression-free survival was 28.8 months with T-DXd versus 6.8 months with T-DM1 (HR 0.33; p<0.000001). The safety profile of T-DXd was manageable.
