Overall survival with ribociclib plus letrozole in advanced breast cancer
Prof. Gabriel Hortobagyi
The multinational phase 3 MONALEESA-2 trial randomized postmenopausal patients with HR-positive, HER2-negative advanced breast cancer to first-line ribociclib plus letrozole or placebo plus letrozole. After a median follow-up of 6.6 years 54.2% of patients in the ribociclib group had died versus 65.6% of patients in the placebo group. The median overall survival was 63.9 months with ribociclib plus letrozole versus 51.4 months with placebo plus letrozole (HR 0.76; two-sided p=0.008). The authors conclude that addition of ribociclib to first-line letrozole among these patients resulted in improvement of median OS by more than one year (quick take video summary).
