Overall survival with pyrotinib plus capecitabine for HER2-positive breast cancer with brain metastases

The multicenter phase 2 PERMEATE trial in China evaluated the combination of pyrotinib and capecitabine among patients with HER2-positive breast cancer with radiotherapy-naïve brain metastases (cohort A) or progressive disease after radiotherapy for brain metastases (cohort B). Overall survival results of the trial have now been published. The median overall survival was 35.9 months (95% CI 24.4-NR) in cohort A and 30.6 months (12.6-33.3) in cohort B. The median CNS-PFS was 13.6 months (95% CI 9.0-15.8) in cohort A and 5.7 months (3.4-11.5) in cohort B. The authors conclude that patients with metastatic breast cancer and brain metastases can reach long-term survival with the combination of pyrotinib plus capecitabine.
