Overall survival with neoadjuvant docetaxel, oxaliplatin, and S-1 followed by surgery and adjuvant S-1 for locally advanced gastric cancer in PRODIGY

The phase III PRODIGY trial, at 18 centers in South Korea, demonstrated that neoadjuvant chemotherapy with docetaxel, oxaliplatin, and S-1 (DOS) followed by surgery and adjuvant S-1 (CSC group) improved progression-free survival compared with surgery and adjuvant S-1 (SC group) for patients with resectable locally advanced gastric cancer (LAGC). Long-term overall survival results of the trial have now been published. With median follow-up duration of 99.5 months, the 8-year OS rate was 63.0% in the CSC group versus 55.1% in the SC group (HR 0.72; p=0.027). With long-term follow-up, CSC also significantly improved PFS (HR 0.70; p=0.016). The authors conclude that neoadjuvant DOS chemotherapy, as part of perioperative chemotherapy, prolonged OS of Korean patients with LAGC relative to patients treated with surgery and adjuvant S-1.
