The Chinese multicenter phase 3 ORIENT-11 study randomized patients with previously untreated locally advanced non-squamous NSCLC to sintilimab (anti-PD-1) plus chemotherapy or placebo plus chemotherapy; with cross-over allowed from placebo to sintilimab at progression. After median 22.9 months of follow-up the median overall survival was not reached in the sintilimab group versus 16.8 months the placebo group (HR 0.60; p=0.0003). Expression of MHC class-II antigen presentation pathway identified patients benefitting most from addition of sintilimab to chemotherapy.
