Overall survival with first-line atezolizumab-vemurafenib-cobimetinib for advanced melanoma with BRAF-V600 mutation

Primary analysis of the phase 3 IMspire150 study showed improved progression-free survival with first-line atezolizumab-vemurafenib-cobimetinib (‘atezolizumab group’) versus placebo-vemurafenib-cobimetinib (‘control group’) among patients with advanced BRAFV600-mutated melanoma with median 18.9 months follow-up (median, 15.1 months versus 10.6 months, HR 0.78; p=0.025). The prespecified second interim analysis of the trial now shows that with median 29.1 months follow-up in the atezolizumab group and 22.8 months in the control group the median overall survival was 39.0 months in the atezolizumab group versus 25.8 months in the control group, a difference that was not statistically significant (HR 0.84; p=0.14). Results of the final analysis are awaited to establish whether a significant improvement in OS can be achieved with long-term treatment with the triplet combination versus vemurafenib plus cobimetinib.
