The multinational phase 3 POLLUX trial randomized patients with relapsed or refractory multiple myeloma (RRMM) to daratumumab-lenalidomide-dexamethasone (D-Rd) or Rd only. Final analysis of the trial found that median overall survival (panel A) and progression-free survival on subsequent line of therapy (panel B) were significantly better in de D-Rd group compared with the Rd group. The OS benefit with D-Rd was seen in most patient subgroups. D-Rd was tolerated. The authors conclude that triplets such as D-Rd are superior to doublets like Rd, and should be standard of care in patients with RRMM.
