Outcomes of pirtobrutinib for relapsed or refractory mantle cell lymphoma in a compassionate us program in Europe

Mantle cell lymphoma (MCL) is a currently uncurable type of B-cell lymphoma. The non-covalent BTK inhibitor pirtobrutinib showed promising response rates in heavily pretreated MCL patients in the approval phase 1-2 BRUIN study, but real-world data are scarce. A retrospective study evaluated pirtobrutinib for relapsed or refractory (R/R) MCL in a compassionate use program in Europe. In this study of 10 patients who had on average three lines of systemic therapy prior to pirtobrutinib, the best overall response rate was 67%. During median 8.6 months follow-up, the median duration of response, progression-free survival, and overall survival were not reached. No new safety signals were observed. The authors conclude that pirtobrutinib was a safe and effective treatment option for R/R MCL in the real world.
