Neoadjuvant trastuzumab, pertuzumab and docetaxel versus trastuzumab emtansine for HER2-positive breast cancer

Dr. Thomas Hatschek

The phase 2 PREDIX HER2 trial, in nine centers in Sweden, randomized patients with HER2-positve breast tumors larger than 20 mm and/or verified lymph node metastases to six cycles neoadjuvant docetaxel, trastuzumab, and pertuzumab (DTP) or trastuzumab emtansine (T-DM1). There was no significant difference between the two groups for the endpoint pCR (45.5% of patients versus 43.9%). Grade 3 or 4 adverse events were seen in 10.1% with T-DM1 versus 39.4% with DTP, including grade 3 or 4 febrile neutropenia in 3.0% versus 26.3%.
