Multinational retrospective cohort study of real-world effectiveness of sacituzumab govitecan for metastatic triple-negative breast cancer

In the ASCENT trial, sacituzumab govitecan (SG; antibody-drug conjugate targeting TROP-2) demonstrated superior efficacy over standard chemotherapy in heavily pretreated metastatic triple-negative breast cancer (mTNBC). Real-world data remain limited. A retrospective study at 18 oncology center in Poland, the Czech Republik, and Slovakia evaluated SG in 303 women with mTNBC. The median follow-up was 8.8 months (IQR 4.5-14.4). The overal response rate was 30.7%. The median progression-free survival and overall survival (co-primary endpoints were 4.4 months and 11.3 months, respectively. The most frequent adverse events were neutropenia (69.0% of patients) and anemia (39.6%). In multivariate analyses, poor ECOG performance status and liver metastases were independently associated with worse outcomes. The authors conclude that this study confirmed the clinical benefit from SG in mTNBC under routine practice conditions.
