Multinational randomized phase 2 study of first-line olaparib plus durvalumab for platinum-ineligible metastatic urothelial carcinoma
Prof. Jonathan Rosenberg
Homologous recombination repair gene mutations (HRRm) are common in urothelial carcinoma, rendering tumor cells sensitive to PARP inhibition. The multinational randomized phase 2 BAYOU trial evaluated addition of olaparib to first-line durvalumab for metastatic urothelial carcinoma (mUC) in platinum-ineligible patients. Addition of olaparib
was not associated with significantly better progression-free-survival in the ITT-population (panel A) but was associated with significantly better PFS in the subset of patients with HRRm (panel B).
