Multinational prospective study of amivantamab plus lazertinib for EGFR-mutant NSCLC after progression on osimertinib plus chemotherapy

Treatment options for patients with EGFR-mutated non-small cell lung cancer (NSCLC) with disease progression on or after osimertinib and platinum-based chemotherapy are limited. Cohort A of the multinational prospective CHRYSALIS-2 study evaluated the combination of amivantamab and lazertinib for these patients. The blinded independent central review-assessed objective response rate was 35% (95% CI 27-42), with median duration of response of 8.3 months (6.7-10.9) and clinical benefit rate 58% (50-66). Duration of response ≥ 6 months was achieved in 57%. Median progression-free survival was 4.5 months (95% CI 4.1-5.8) and median overall survival was 14.8 months (12.2-18.0).Grade ≥3 adverse events were rash (10% of patients), infusion-related reaction (9%), and hypoalbuminemia (6%). The authors conclude that among patients with EFGR-mutated NSCLC and progression on or after osimertinib plus chemotherapy, the combination of amivantamab plus lazertinib had antitumor activity with a safety profile that was generally manageable (graphical abstract ).
