The multinational phase IIb ADAGIO study evaluated the selective Wee1 inhibitor adavosertib (AZD1775) for recurrent or persistent uterine serous carcinoma (UCS) after platinum-based chemotherapy. Adavosertib was administered orally at 300 mg once daily on days 1-5 and 8-12 of 21-days cycles. The objective response rate was 26.0% (95% CI 17.9-35.5) with median duration of response 4.7 months (3.8-8.3), median progression-free survival 2.8 months (2.6-3.9) and median overall survival 9.6 months (8.3-NC). Most patients (97.2%) experienced treatment-related adverse events, with grade ≥ TRAEs in 60.6% and TRAEs leading to discontinuation in 14.7%. The authors conclude that adavosertib had some antitumor activity in patients with recurrent or persistent USC, but was not well tolerated at 300 mg once daily dosing.
